Function

During the mid- to late phase of seed development, the embryo and the aleurone tissue undergo a maturation process, whereby each becomes dormant or quiescent and acquires the ability to tolerate desiccation. The maturation process is associated with the accumulation of characteristic sets of mRNAs and proteins, which are products of the so-called LEA (Late Embryogenesis Abundant protein) genes (Baker et al. 1988, Skriver and Mundy 1990). The phytohormone ABA plays key roles in the control of these events (McCarty 1995, Leung and Giraudat 1998). Genetic loci have been identified in Arabidopsis and maize that are required for the control of maturation processes. The viviparous1 (vp1) mutant of maize has been characterized as an ABA-insensitive mutant, in which various events during seed maturation are blocked (McCarty et al. 1991, Robichaud et al. 1980). At the molecular level, the vp1 mutant fails to accumulate mRNAs of ABA-regulated LEA genes, such as Rab28 (Pla et al. 1989) and the maize homologue of the wheat Em gene (McCarty et al. 1991), as well as the genes for globulin storage proteins (Kriz et al. 1990). The Vp1 gene encodes a transcription factor that can activate the transcription of maturation-associated genes(McCarty et al. 1991, Hattori et al. 1992). In Arabidopsis, ABI3, one of the genes required for ABA response in seeds is known to be orthologous to maize VP1 (Giraudat et al. 1992). Mutant seeds with severe abi3 alleles show vivipary and block the expression of LEA and storage protein genes (Parcy et al. 1994). Parcy et al. (1994) have shown that transgenic Arabidopsis ectopically expressing ABI3 can induce expression of seed-specific genes in non-seed tissues in response to ABA. Thus, VP1/ABI3 is considered to function in determining the seed specificity of certain sets of ABA-regulated genes. Homologues of VP1/ABI3 have also been cloned from several plant species such as tobacco (Phillips and Conrad 1994), bean (Bobb et al. 1995), and wild oat Avena fauta (Jones et al. 1997). We have cloned the rice orthologue of VP1 (OSVP1) and have been studying the mechanism of tissue/organ-specific regulation of ABA-responsive genes such as the rice Em (OSEM) gene, in which OSVP1 is expected to play a key role (Hattori et al. 1994, Hattori et al. 1995). VP1/ABI3 functions as a transcription factor by acting on at least two distinct types of cis-elements, namely, ABA-response elements (ABREs) and RY/Sph elements. While VP1/ABI3 acts on ABREs via the interaction with ABRE-binding factors such as TRAB1 and ABI5 (Hobo et al. 1999a, Finkelstein and Lynch 2000, Lopez-Molina and Chua 2000, Nakamura et al. 2001), it directly binds to the RY/Sph element through its own DNA binding domain called the B3-domain (Suzuki et al. 1997).

Expression

Temporal and spatial regulation of OSVP1 expression during embryo development Temporal and spatial patterns of OSVP1 expression, at both the mRNA and protein level, were examined by in situ hybridization and immuno-localization analyses. OSVP1 mRNA was not detected in the embryo at 1 DAP (Fig. 1A), but was expressed in the whole embryo by 2 DAP (Fig. 1B). Examination of multiple sections from independent seeds revealed that OSVP1 mRNA became detectable in the embryos of 50–70 µm in length, which corresponded to the mid-globular stage. Until 4 DAP, when the coleoptiles started to differentiate, OSVP1 mRNA was distributed uniformly throughout the embryo (Fig. 1B–D). At 6 DAP, however, the signals became localized to those regions where the shoot and radicle were developing (Fig. 1E). A clearer distribution of OSVP1 mRNA could be observed when all the embryonic organs were fully established (Fig. 1F–H). OSVP1 mRNA could be detected in the vascular tissues, a part of coleoptiles, leaves, shoot meristem, and radicle. Epidermal layers of the scutellum and epiblast also showed stronger signals than the inner tissues of these organs. Although other parts of the embryo appeared to express lower levels of OSVP1, it was not certain whether those signals were above the non-specific background. Overall levels of OSVP1 mRNA appeared to decrease by the mature 20 DAP stage (Fig. 1H) compared to those at 10–15 DAP (Fig. 1F, G). In the endosperm, OSVP1 mRNA was detected only in the aleurone layers, the outermost cell layers, after 6 DAP when they had just differentiated, but not in the inner tissues (Fig. 2A–D).
