Annotated Information

Function

OsVIT1 and OsVIT2 may function primarily in flag leaves to mediate vacuolar sequestration of Fe and Zn, due to highly expression levels of OsVIT1 and OsVIT2 in flag leaves rather than embryos. And they might also mediate allocation of Fe/Zn between flag leaves and the grains[1]. Knockdown of the vacuolar Fe transporter gene OsVIT1 or OsVIT2 may be useful to further increase the Fe concentration of seeds [2].

Expression

OsVIT2 locates in chromosome 4 with four exons and three introns as predicted, and encodes putative protein as the vacuolar iron transporter with 252 amino acids which shares 80.6% amino acid similarity with the VIT in Arabidopsis. There are five conserved transmembrane domains in OsVIT1 by further analyses. This gene expresses in flag leaf blades with a significantly high level[1]. different from OsVIT2 was quickly induced in rice seedling roots and shoots after the treatment of 4.5 mM FeSO4, OsVIT1 was not highly induced. Fe starvation caused downregulated OsVIT2 expression in roots and shoots, and no OsVIT1 response until 7 days after treatment in roots. These data suggest that OsVIT1 may represent a housekeeping mechanism for Fe accumulation in vacuoles, while OsVIT2 functions more in response to external Fe levels[1].

Subcellular localization

Figure 1.Subcellular localization of OsVIT1 and OsVIT2[1].(a–c) Bright-field images of Arabidopsis protoplasts expressing enhanced green fluorescent protein (EGFP) (a), OsVIT1: EGFP (b) and OsVIT2: EGFP(c).(d–f) Fluorescence images of Arabidopsis protoplasts expressing EGFP (d), OsVIT1: EGFP (e) and OsVIT2: EGFP(f).(g–i) Merged fluorescence images of EGFP (g), OsVIT1: EGFP (h) and OsVIT2: EGFP (i). Bars = 10μm.

OsVIT1 and OsVIT2 were localized to vacuolar membrane[1] when they combined with enhanced green fluorescent protein and expressed(Figure 1).
