(A) and (B) Wild-type (WT) and rss3 seedlings were grown with (+) or without (−) 100 mM NaCl for 7 d.

(A) Seedlings.

(B) Roots floating on water.

(C) Propidium iodide staining of the primary roots of 3-d-old wild-type and rss3 seedlings, grown in the presence or absence of 100 mM NaCl. Red and white arrowheads indicate the border of the MZ and EZ, and of the EZ and MTZ, respectively. Images were generated by concatenating two or more pictures.

(D) Length (left) and number of cells (right) in the MZ and EZ of the root tips of 3-d-old wild-type and rss3 seedlings, grown with or without 100 mM NaCl. Mean ± sd, n ≥ 3. Asterisks indicate a Student’s t test P value < 0.05.

(E) The MTZs of 7-d-old roots of wild-type and rss3 seedlings grown in the presence or absence of NaCl were observed by differential interference contrast microscopy.

Bars = 5 cm in (A), 1 cm in (B), 400 µm in (C), and 100 µm in (E).
