WOX3, a rice WUSCHEL-LIKE HOMEOBOX gene, negatively regulates the rice YAB3 gene which is involved in the leaf development in rice.

Annotated Information

Function

WOX3 funtions as a transcriptional repressor of YAB3.

The YABBY family encode transcription factors with a C2C2 zinc finger domain toward the amino terminus and a putative helix-loop-helix domain called YABBY. YABBY members in Arabidopsis function in controlling abaxial identity of lateral organs, but funtion differently in monocots, with an example of maize FIL-related YABBY genes(ZmYAB9 and ZmYAB14) expressing on the adaxial part of leaf primordia indicating a probable role in lateral leaf outgrowth rather than specifying adaxial cell fate. Likely, rice YABBY genes have different funtions from YABBY members in Arabidopsis and in maize. Additionally, these rice YABBY genes have distinct funtiions. In particular, YAB3 contributes to the exclusion of KNOX expression from the leaves(KNOX genes are essencial for leaf development), that is YAB3 is required for the promotion of diffentiation during leaf organogenesis.

Transgenetic experiments show that overexpression of WOX3 represses YAB3 and repression of WOX3 by RNAi induces the expression of YAB3. Further sequence analysis and expression and DNA-binding studies showing WOX3 can actually bind to the WOX3-binding site in YAB3 reinforces the hypothesis that WOX3 has a function to control or limit the expression levels of YAB3 in leaves contributing to maintain the undifferentiated state of dividing cells.

In summary, a regulatory module of rice leaf development in which WOX3 represses YAB3 that in turn represses KNOX genes in leaf tissues is identified. And this regulatory relationship among these three genes might be required to maintain the balance between cell division and cell differentiation during the leaf growth.

Expression

WOX3 and YAB3 are coexpressed in most lateral organ primordia and young leaves without adaxial/abaxial polarity.

Overexpression of WOX3 leads to lnotted outgrowth of leaves that lack clear separation between the leaf sheath and the leaf blade, simiar to thephenotypic changes as induced by YAB3 RNAi, along with ectopic expression of KNOX genes in leaves of the trangenic plants. These experiments with inducible WOX3 expression suggest that WOX3 directly regulated YAB3. Conversely, down-regulation of WOX3 induces no phenotype which suggests that other WOX genes may act redundantly with WOX3 to function independently of YAB3/KNOX regulation. Evolution

According to the phylogeny analysis of YABBY and WOX families, OsWOX3 is highly homologous to Arabidopsis PRS gene and the maize dulicated NS1 and NS2 genes, while OsYAB3 falls to a small FIL/YAB subgroup consisting of AtYAB3, StYAB, FIL, ZmYAB14, and OsYAB3. Knowledge Extension

Glabrous Rice 1(GRL1) shares the same gene locus with WOX3. Sequence analysis showed that GRL1 is similar to OsWOX in rice. GRL1 encodes WUS-like homeodomain protein and is essential for the trichome development in rice and will contribute to breeding of glabrous elite rie varieties.Figure1.jpg
