Predicting therapeutic drugs for hepatocellular carcinoma based on tissue specific A new approach for predicting therapeutic drugs for HCC based on tissue-specific pathways and identified three newly predicted drugs that are likely to be therapeutic drugs for the treatment of HCC.

Introduction

A novel approach was proposed to identify related drugs for the treatment of HCC, which considers the functional information of pathways, the changes of a single gene in pathways, and the special relationship between HCC and liver tissue. The up-regulated and down-regulated genes were searched  in each cancer-associated pathway that was counter-regulated the most by each drug instance in the Connectivity Map (CMap)  database. The degree of counter-regulation of those two kinds of gene was combined for each drug instance. The combined degrees were averaged over all cancer-associated pathway genes and instances of each drug, giving one therapeutic score for each drug. Drugs were ranked by the therapeutic scores, and top ranked drugs selected for further investigation.
