Description We propose a novel sequencing protocol for circular RNA enrichment and full-length sequencing of circular RNA isoforms using Nanopore sequencing. By utilizing circular reverse transcription and fragment size selection, circRNAs can be efficiently enriched (20 fold higher than previous methods) from total RNA. With the advantages of Nanopore sequencing, our method has promising applicability in unbiased characterization and reconstruction of full-length circRNA sequences.
