| 描述信息 | Human papillomavirus (HPV) is an important pathogenic agent of cervical cancer. The integration frequency and integration site of different HPV strains in human genome are different in the risk of cervical cancer. HPV encoded oncogenes E6 and E7 lead to a large number of genomic structural mutations through interfering WITH DNA repair, which are directly related to the occurrence and development of cervical cancer. Currently, the second-generation whole genome sequencing widely used in HPV detection is difficult to accurately analyze integration sites and structural variations. Nanopore third-generation sequencing (ONT), with a single read length of 1Mb, can make up for the shortcomings of second-generation sequencing. The project team has collected 101 cervical cancer tissue samples and clinical case data. The third-generation sequencing of two cervical cancer patients was completed in the early stage, and 160 integration sites were found in one high type HPV35, and 70 genes were inserted in ADORA1, PIK3C2A and so on. We will further analyze and verify whether HPV genomic insertion affects the expression of genes at the insertion site and the expression of related pathway proteins regulated by these genes. Another low - risk HPV82 integration site 25, the results need further analysis. In addition, the project plans to develop integration sites and SVs analysis and visualization software for clinical detection and target prediction of precision treatment of cervical cancer. This study will fill in the gap of three generations of sequencing in the accurate diagnosis of cervical cancer, and is expected to provide a new analysis platform for cervical precision therapy. | ||||||||
