Small cell carcinoma of the esophagus (SCCE) is a rare and aggressive malignancy that poses significant clinical challenges. This study employs single-cell RNA sequencing (scRNA-seq) to investigate SCCE samples from a single patient before and after chemotherapy. The analysis reveals notable cellular plasticity and changes in the tumor microenvironment's cellular composition, including increased tumor cell diversity and decreased populations of T cells, B cells, and myeloid-like cells. Pre-treatment samples predominantly show carcinoma cells in a middle transitional state, while post-treatment samples exhibit expanded populations of cells in terminal and universally altered states. Dynamic interactions between tumor and immune cells, particularly through key signaling pathways such as TIGIT-PVR and MDK-SDC4, are highlighted. This research offers new insights into the adaptive mechanisms of SCCE following chemotherapy and identifies potential therapeutic targets to enhance treatment efficacy. |
