Here, we profiled the single-cell transcriptomes of 13,289 peripheral blood mononuclear cells isolated at three longitudinal stages from two severe COVID-19 patients treated with Tocilizumab. We identified a severe stage-specific monocyte subpopulation that contributed to patients' inflammatory storm. Further investigation of the other differential cell subtypes revealed a boosted humoral and cell-mediated immunity to respond viral infection in severe COVID-19 patients. Finally, we illustrated a severe stage-specific monocyte-centric immune cell interaction network connected by the inflammatory cytokines and their receptors. These results provided additional insights into the immunopathogenesis of COVID-19 and candidate drugs for the treatment of severe patients. |
