HRA001740  (Open Access) DNA methylation, as a crucial epigenetic modification, plays an essential role in genome regulation. Dysregulation of DNA methylation usually means disruption of development and disease.Here, we developed carrier-assisted base-conversion by enzymatic reaction with end-tagging (Cabernet) technology, which enables bisulfite-free detection of 5-methylcytosine (5mC) and 5-hydroxymethylcytosine (5hmC) at single-cell level and single-base resolution. Cabernet can provide a high-detectability method for 5mC, 5hmC and hemi-methylation profiling at single-cell level.
