Extrachromosomal DNA is associated with the chromothripsis event and the diverse patient prognosis in the gastric cardia adenocarcinoma

In this study, we investigated the availability and function of extrachromosomal DNA in the gastric cardia adenocarcinoma (GCA) from a Chinese cohort of gastric cardia adenocarcinoma with whole-genome sequencing (WGS), whole-exome sequencing (WES), and immunohistochemistry. For the first time, we identified the ecDNA amplicons present in most GCA patients, and found that some oncogenes are present as ecDNA amplicons in these patients. These oncogene ecDNA amplicons are associated with the chromothripsis process. Strikingly, we found that two oncogenes amplicons have a diverse correlation with patients prognosis. The diverse association of ecDNA amplicons and patient prognosis may help us to design personal therapy strategies for these GCA patients.
