Deciphering early human pancreas development at the single-cell level

Here, we performed scRNA-seq of human embryonic pancreas tissues collected from PCW 4 to 11 and scATAC-seq from PCW 8 to 11. We profiled the major cell types of pancreatic epithelial cells, including dorsal and ventral MP cells, and revealed their molecular heterogeneity and developmental trajectory. We analyzed the transcription factors, regulatory networks and signaling pathways of acinar and ductal lineage cells and revealed a regulatory network of endocrinogenesis and transcription dynamics of EP and endocrine cells. We further compared the developmental trajectories between humans and mice and demonstrated a few distinct features, such as gene expression patterns, in each species. Taken together, our data depict the whole progression of pancreatic organogenesis during the first trimester (PCW 4-11) at the single-cell level.
