Biallelic inactivation of TP53 and elevated copy-number alterations promote clonal expansion at early stage of esophageal carcinogenesis

Description:
We conducted whole-genome and whole-exome sequencing of LCM-dissected micro-biopsies from 4 esophageal squamous cell carcinoma (ESCC) patients, 10 individuals with pre-cancer intraepithelial neoplasia, and 14 individuals without cancer or pre-cancer pathology from a ESCC high-risk region in China. For each sample, multiple adjacent normal and neoplasia micro-biopsies were collected and sequenced, enabling us to reconstruct the phylogeny and reveal the dynamic mutational landscape during the cancer development. Our results delineated a clear evolutionary path from clones in normal aging esophagus to ESCC.
