Fostering the Transition to Human Ground-State Pluripotency through Precise NFkB/RELA Circuitry Inhibition

Description:
The transition from human primed pluripotency to a naive state offers insight into embryo implantation. We developed a serum-free system, 6iLV, achieving a 90% conversion rate in 12 days. Multi-omics sequencing revealed chromatin accessibility changes, with increased openness at OCT and KLF binding motifs and decreased accessibility at ZIC, RFX, and NFkB/RELA motifs. MicroRNA profiling showed elevated miR-372/373 and miR-182/183 clusters and decreased miR-302 and miR-363/106 cluster expression. Functional experiments demonstrated that miR-372/373 overexpression promoted the transition, while NFkB/RELA overexpression inhibited it. Mechanistically, miR-372/373 suppressed the expression of NFkB/RELA, facilitating the transition.
