Description:
To investigate the molecular heterogeneity and dynamic responses of acute myeloid leukemia (AML) subpopulations, we performed a controlled ex vivo perturbation study followed by transcriptional profiling. Peripheral blood or bone marrow samples were collected from de novo AML patients (n=20, diagnosed per WHO criteria). Mononuclear cells were isolated by density gradient centrifugation and subjected to fluorescence-activated cell sorting (FACS) to separate two phenotypically distinct subsets: SMALL cells and LARGE cells. Each purified subset was allocated to experimental arms with specific pharmacological interventions:SMALL cell cohort: Untreated control (n=5) vs. Jasplakinolide (Jas, 100 nM, 6h) treatment (n=5); LARGE cell cohort: Untreated control (n=5) vs. Latrunculin A (Lat A, 5 uM 6h) treatment (n=5); Following 6-hour incubation under standardized culture conditions (37°C, 5% CO2), all four groups (SMALL, SMALL+Jas, LARGE, LARGE+Lat A) were processed for bulk RNA sequencing.
