The embryos were lysed directly and used for cDNA synthesis using SMART-Seq v4 Ultra Low Input RNA Kit for Sequencing (Takara, 634888). The cDNA samples were fragmented to 200-400bp by Covaris sonicator (Covaris). Sequencing libraries were prepared with NEBNext Ultra II DNA Library Prep Kit for Illumina (NEB, E7645S). |
