Nuclear DNA from young leaf was cross-linked in situ, extracted, and then digested with a restriction enzyme (MboI). The sticky ends of the digested fragments were biotinylated, diluted, and then ligated to each other randomly. Biotinylated DNA fragments were enriched and sheared again for preparing the sequencing library, which was sequenced on the Illumina HiSeq X Ten platform |
