To comprehensively expound the adaptive changes of liver during transition period, liver biopsies were conducted twice on 20 transition cows at average 7d before calving and average 16d after calving. Transcriptome profile of the liver samples were detected using RNAseq. Accurate 3 μg RNA per sample was utilized as input for complementary DNA (cDNA) libraries construction. Sequencing libraries was generated using the rRNA-depleted RNA following manufacturer’s recommendations, with NEBNext® Ultra™ Directional RNA Library Prep Kit for Illumina® (NEB, USA). The RNA sequencing was performed on an Illumina HiSeq 4000 platform at the Novogene Bioinformatics Institute (Beijing, China) according to the manufacturer’s instructions. |
