Total DNA of the four development stages (blastula period, gastrula period, segmentation period and hatching period) of the allotetraploid lineage (RCC, CC, F1 allodiploid and F22 allotetraploid) were extracted using a QIAamp DNA Mini Kit (Qiagen, Chatsworth, CA, USA), and then was fragmented with a Covaris S2 Ultrasonicator. DNA methylated library of each sample was constructed using a Whole Genome Bisulfite Sequencing (WGBS) library prep kit following the manufacturer’s instructions. |
