Cells were lysed with lysis buffer supplemented with protease inhibitor cocktail and 1 mM PMSF (1x final each) and then sonicated to fragments about 200-500 bp using a Bioruptor. Immunoprecipitated DNA was isolated with H3K27ac antibodies (Abcam, Cat.N: ab4729). Immunoprecipitated DNA was used to construct sequencing libraries following the protocol provided by the NEXTflex™ ChIP-Seq Kit (Bioo Scientific, Cat.N: NOVA-5143-02). All libraries were sequenced on HiSeq X Ten (Illumina) platform to produce an average of ~120.62 million 150-bp paired-end raw reads and ~115.78 million high-quality reads for each library. |
