Fresh leaves were fixed in NIB buffer. Cross-linked DNA was digested with HindIII, followed by biotinylated nucleotides for filling and proximity ligation. After reverse cross-linking and purification, DNA was sheared to 300-700 bp and five Hi-C libraries were prepared. |
