For ULI-MNase-seq, 10-15 cells were collected per reaction, and at least two replicates were performed for each stage. The cells were isolated and washed thoroughly in 0.5% BSA-PBS in order to avoid pollution. The ULI-MNase libraries were generated according to a pervious protocol. The sequencing libraries were prepared using the KAPA Hyper Prep kit for the Illumina platform (kk8504) following the manufacturer’s instructions. |
