Total faecal DNA was extracted from the supernatant using QIAamp DNA Stool Mini Kit (Qiagen). Metagenomic DNA of samples were sheared into ~300bp fragments using a focused ultrasonicator (Covaris, Woburn, MA). Sequencing libraries were constructed with the sheared fragments using TruSeq Sample Preparation Kit v2 (Illumina). |
