The rRNA was removed from all samples with RINe > 8.0 and and no less than 2 μg. RNA purification, cDNA library construction and RNA sequencing were conducted by Berry Genomics company. Specifically, the NEBNext® UltraTM Directional RNA Library Prep Kit was used for Illumina® to make rRNA-free RNA sequencing libraries. Then, the firststrand cDNA was synthesized using random hexamer primers and M-MuLV reverse transcriptase (RNaseH-), and cDNA second strand synthesis was performed using DNA polymerase I and RNase H. Each library was paired-end sequenced (2×150 bp) on an Illumina Novaseq6000 platform. |
