Fresh young leaf tissue was collected from Bst99 plants. We constructed SMRTbell libraries using a method described in a previous study. Genomic DNA was isolated and sheared by Megaruptor, followed by damage repair, end repair, ligation with known adapters, enzyme digestion, size-selection, and generation of SMRTbell structure library. The qualified libraries were sequenced on the PacBio Sequel II platform (Pacific Biosciences) at BGI-ShenZhen Company, generating approximately 19.4 Gb of PacBio HiFi reads for the Bst99 genome.
