Similar to NGS library preparation, scRNA library and VDJ library preparation were also performed by DNA fragmentation, end repair, A-tailing, adaptor ligation and sample index PCR. The post ligation cleanup cDNA was amplified using the following program: 98 °C for 45s, followed by 15 cycles of 98 °C for 20 s, 67 °C for 30 s and 72 °C for 20s. 1 µl of sample (1:10 dilution) was added to an Agilent Bioanalyzer High Sensitivity chip(Agilent, 5067-4626) to determine the average fragment size. Library concentration was determined using the Qubit dsDNA HS Assay Kit(Thermo, Q32854) |
