Total RNA of fungi was extracted according to the instructions of RNA kit. The concentration, purity and integrity of RNA in the samples were detected by low light level spectrophotometer. Qualified RNA samples will be tested for cDNA library construction, library quality detection and high-throughput sequencing. The mRNA is enriched by removing rRNA, and the enriched mRNA is reversely transcribed to form double-stranded cDNA, the double-ends and splices of cDNA are repaired, and then PCR amplification is performed to construct cDNA library. |
