Nuclei were extracted from the sample after homogenization, and bound to Concanavalin A-coated magnetic beads, followed by the incubation with the Abcam H3K4me3 Rabbit pAb. Nuclei were then successively incubated with the ABclonal Goat anti-Rabbit IgG H&L, pA-Tn5 adapter complex preparation, and tagmentation buffer, followed by Proteinase K treatment to stop tagmentation. DNA was extracted using the Beckman Coulter AMPure XP beads, and purified using the AMPure XP beads after the amplification with the NEBNext HiFi 2× PCR Master Mix, universal i5 primer and uniquely barcoded i7 primer. Library was assessed by the Life Invitrogen Qubit 3.0 and the Agilent Bioanalyzer 2100 system. |
