melon genomic DNA was sheared to 20 kb using G-tubes, then purified, damage repair, hairpin adaptor ligation, and digestion with exonuclease to remove the damaged DNA and fragments that without adaptors. The target fragments were size-selected by Blue Pippin electrophoresis.
