Whole genome bisulfite sequencing libraries were constructed using the Acegen Bisulfite-Seq Library Prep Kit according to the manufacturer's protocol. Briefly 1 μg of genomic DNA spiked with 1 ng unmethylated Lambda DNA was fragmented by sonication to a mean size of approximately 200-500 bp. After bisulfite treatment the DNA was amplified with 10 cycles of PCR using Illumina 8-bp dual index primers. The constructed WGBS libraries were then analyzed by Agilent 2100 Bioanalyzer and finally sequenced on Illumina platforms using a 150*2 paired-end sequencing protocol. |
