RNA was extracted from tissue samples using the NanoDrop 2000 and treated with DNase I (RNase-free). A total amount of 5 μg RNA per sample was used as input material for RNA-seq library preparation. Sequence libraries were generated using NEBNext Ultra RNA Library Prep Kit and index codes were added to attribute sequences to sample. To select cDNA fragments of preferentially 250-301 bp in length, the library fragments were purified with AMPure XP system. |
