total RNA samples were extracted from mettle-depleted mouse B cells using Trizol reagent and then subjected to small RNA purification using mirVana miRNA Isolation Kit. The isolated small RNAs were demethylated with recombinant wildtype AlkB and D135S AlkB proteins. The demethylated small RNAs underwent cDNA library construction using the NEBNext Multiplex Small RNA library Prep Set for illumina Kit. High throughput sequencing were performed on these libraries. |
