total RNA samples were extracted from wildtype mouse B cells using Trizol reagent and then subjected to small RNA purification using mirVana miRNA Isolation Kit. The isolated small RNAs were demethylated with recombinant wildtype AlkB and D135S AlkB proteins. The demethylated small RNAs were treated with 0.2M NaBH4 on ice for 30 min in the dark and then purified using the Oligo Clean & Concentrator Kit. The NaBH4-treated small RNAs were further incubated with 100mL cleavage buffer (H2O:glacial acid:aniline=7:3:1) at RT in dark for 2 h. After purification using the Oligo Clean & Concentrator Kit, the small RNAs underwent cDNA library construction using the NEBNext Multiplex Small RNA Library Prep Set for illumina Kit. High-throughput sequencing were performed on these libraries. |
