Qualified DNA of young healthy leave samples are randomly fragmented using a Covaris ultrasonic disruptor. Enrichment and purification of large DNA fragments are performed using magnetic beads. Fragmented DNA is subjected to damage repair and terminal repair; Circular sequencing adapters are connected at both ends of the DNA fragment, and failed segments are removed using exonuclease. The constructed library is sequenced using Pacbio Sequel IIe. |
