Timed samples with two duplicates were collected every 4 hour for 48 hr under LD condition, each sample from three individuals. A total amount of 3 g RNA per sample was used for the RNA sample preparations. Sequencing libraries were generated using NEBNextUltra RNA Library Prep Kit (NEB, USA) following the manufacturers instructions. The clustering of the index-coded samples was performed on a cBot Cluster Generation System using TruSeq PE Cluster Kit v4-cBot-HS (Illumia) according to the manufacturers instructions. After cluster generation, the library preparations were sequenced on an Illumina platform and paired-end reads were generated. |
