For Hi-C sequencing, formaldehyde was used for crosslinking the fresh young flower buds. DNA were digested with the restriction endonuclease Hind III. DNA fragments were labeled with biotin through end repair. The purified DNA fragments were circularized and broken into a length of 300-700 bp. Finally, the DNA fragments captured by streptavidin protein beads were used for Hi-C library construction. |
