Similar to NGS library preparation, scRNA library and VDJ library preparation were performed by DNA fragmentation, end repair, A-tailing, adaptor ligation and sample index PCR. The post ligation cleanup cDNA was amplified. 1 ul of sample (1:10 dilution) was added to an Agilent Bioanalyzer High Sensitivity chip(Agilent, 5067-4626) to determine the average fragment size. Library concentration was determined using the Qubit dsDNA HS Assay Kit(Thermo, Q32862) |
