Description | HSC were sorted from femur bone marrow of 2D2 TCR (TCR-MOG) transgenic and wild type mice. Cells were directly sorted into Trizol LS (ThermoFisher)-containing tubes, and total RNA was then extracted. Each mRNA was hibridized with two ~50 base probes in the hybidize solution. The reporter probe carries the signal, the capture probe allows the complex to be immobilized for data collection. After hybridization, excess probes are removed and the probe/target complexes aligned and immobilized in the nCounter Cartrige. Sample cartriges are placed in the Digital Analyzer. Color codes on the surface of the cartrige are counted and tabulated for each target mRNA. Data were processed on the nSolver software. Data were normalized to the 14 reference genes including positive and negative controls and housekeeping genes. |
