| Description | Sample Preparation Serum samples stored at -80 were thawed at room temperature.150 μL of sample was added to a 1.5 mL Eppendorf tube. Subsequently, 600 μL of ice-cold mixture of methanol and acetonitrile (2/1, vol/vol) was added, and the mixtures were vortexed for 1 min, and the whole samples were extracted by ultrasonic for 10 min in ice-water bath, stored at -40 for overnight. The extract was centrifuged at at 4°C (12000 rpm) for 10 min. 150 μL of the supernatant was aspirated with a syringe, filtered through a 0.22 μm organic phase pinhole filter, transferred to an LC injection vial, and stored at -80 until LC-MS analysis was performed. QC samples were prepared by mixing aliquot of the all samples to be a pooled sample. |
