Title scRNA-seq and spatial transcriptomics for direct tumoral interactome
Description We applied TurboID-mediated intercellular labeling (STCL) to characterize the dynamic tumor-epithelium interaction using a tumor-suppressive cell competition mouse model, and performed the interaction-based transcriptome analysis by scRNA and spatial transcriptome. We first subcloned MFC-STCL cells and finally obtained an orthotopic tumor-suppressive GC model, in which the tumor size was gradually increased from day 0 to day 10 but eventually diminished at day 15. Then we harvested the whole stomach (including tumors) at day 5 (early), day 10 (middle), and day 15 (late), digested to obtain single-cell suspension for flow cytometry analysis to discriminate biotin+ (interacting) and biotin- (bystander), and processed for scRNA to analyze the interacting cellular components. We also subjected the stomach (tumor) tissue collected at indicated time points to spatial transcriptomics analysis to gain insights of the spatial distribution of molecular events.
