Monocytes were isolated from the peripheral blood mononuclear cells (PBMCs) of 10 healthy donors via magnetic activated cell sorting (MACS) with Human CD14+ MicroBeads. Subsequently, these monocytes were differentiated into monocyte - derived macrophages (MDMs) through incubation with 50 ng/ml M-CSF for seven days. Following this, the MDMs were exposed to oxLDL to induce the formation of foamy macrophages (FMs). Both the human MDMs and FMs were then infected with M. leprae. Total RNA was extracted from these two groups of cells and subjected to transcriptome sequencing using the Illumina Novaseq 6000 platform. |
