8-week- old male C57BL/6 mice (8 weeks old) were administrated with 30 days of high-fat diet (HFD) feeding, streptozotocin (STZ) was dissolved in citrate buffer (pH 4.2) at a concentration corresponding to 50 mg/kg body weight. Mice were intraperitoneally injected with STZ once daily for 5 consecutive days; Seven days later, mice with a fasting blood glucose level > 11.1 mmol/L were considered successfully modeled. Control mice received intraperitoneal injections of an equal volume of citrate buffer alone under the same experimental conditions. Then, mice were anesthetized and perfused with PBS. Then, liver tissues were harvested and shipped to Guangzhou Gene Denovo Biotechnology Co., Ltd. for sample dissociation and further sequencing. |
