Description | The aim of the untargeted metabolic profiling study is to obtain a global metabolome coverage from biological samples. Therefore, a comprehensive and systematic protocol for tissue metabolite extraction is highly desirable. In this study, we evaluated a comprehensive liver pretreatment strategy based on ultra-high-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF-MS) to expand metabolome coverage using four different protocols. These protocols included (A) methanol (MeOH) protein precipitation, (B) two-step extraction of dichloromethane (DCM)/MeOH followed by MeOH/H2O, (C) two-step extraction of methyl tert-butyl ether (MTBE)/MeOH followed by MeOH/H2O, and (D) two-step extraction of isopropanol (IPA)/MeOH followed by MeOH/H2O. |
