Description | We report that TAM secretes abundant pleiotropic protein (PTN) to stimulate glioma stem cells (GSC) through its receptor PTPRZ1, thereby promoting the malignant growth of GBM through PTN-PTPRZ1 paracrine signaling. The expression of PTN is correlated with the infiltration of CD11b+/CD163+ TAM and the poor prognosis of GBM patients. Co-implantation of M2-like macrophages (MLC) promotes GSC-driven tumor growth, but silencing PTN expression in MLC reduces their pro-tumorigenic activity. The PTN receptor PTPRZ1 is preferentially expressed in GSC and also predicts poor prognosis for GBM. Destruction of PTPRZ1 abolished the maintenance and tumorigenic potential of GSC. |
