Description | The cells were lysed and extracted using a PierceTM Classic Magnetic IP/Co-IP kit (Thermo Fisher Scientific, Waltham, MA, USA) and subjected to IP using 20 µg of YY1 antibodies. The precipitated protein complex was separated using sodium dodecyl sulfate polyacrylamide gel electrophoresis and subjected to silver staining according to the manufacturer's instructions (Thermo Fisher Scientific). After band excision and peptide extraction and digestion, the digest was subjected to HPLC-MS analysis using the EASY-nLCTM 1200 UHPLC system (Thermo Fisher Scientific) on an Orbitrap Q Exactive HF-X mass spectrometer (Thermo Fisher Scientific). Each fraction was analyzed using Proteome Discoverer 2.2 (Thermo Fisher Scientific). |
