Function
- Actin-interacting protein 1 (AIP1) has been shown to be an important factor by acting coordinately with actin-depolymerizing factor (ADF)/cofilin in promoting actin depolymerization, the rate-limiting factor in actin turnover.
- Specific down-regulation of OsAIP1 increased the level of filamentous actin and reduced actin turnover, whereas over-expression of OsAIP1 induced fragmentation and depolymerization of actin filaments and enhanced actin turnover.
- OsAIP1 acts coordinately with ADF and profilin to induce massive net actin depolymerization, indicating that AIP1 plays a major role in the turnover of actin, which is required to optimize F-actin levels in plants.
Phenotypic analysis
- The actin cytoskeleton appeared to form a uniformly distributed network in control pollen grains (Figure 2a), but appeared fragmented in OsAIP1 over-expression pollen grains (Figure 2b), suggesting that OsAIP1 over-expression induced actin depolymerization.
- By contrast, actin filaments became denser and more highly bundled in OsAIP1 RNAi pollen grains (Figure 2c). The F-actin level was decreased in OsAIP1 over-expression pollen grains but increased in OsAIP1 RNAi pollen grains (Figure 2d), and the F-actin level inversely correlated with the amount of OsAIP1 transcript, suggesting that OsAIP1 inhibits actin polymerization.
