- The researchers generated two fusion proteins in which green fluorescent protein (GFP) was terminally fused with either wild-type OsVPE1 (pNVG) or mutant OsVPE1 (pWVG). These proteins were expressed in onion epidermal cells. As expected, the control GFP signals localize to both the cytoplasm and nuclei of the cells (Figure 8a). Fusion proteins expressed from both the pNVG and pWVG constructs were detected in vacuoles (Figure 8b,c).

- Multiple sequence alignment shows that this VPE is more closely related to several previously identified seed-type VPEs (such as Arabidopsis bVPE, castor bean (Ricinus communis L.) VPE, rice NP1 and rice REP2) than to vegetative-type VPEs (such as AtaVPE, cVPE, dVPE and Proteinase B) (Figure 4).
