Function

Os06g0254600 is a stress responsive gene. It encodes the caleosin which mainly lay in oil body or lipid body. Caleosins not only promote formation of oil body but also is related to drought stress.

Expression

It includes six exons and the length of the second intron is 5kb.It is located on chromosome 6.There are two other caleosin genes-Os06g0254300 and Os06g0254700 in chromosome 6.These three genes get together because tandem duplication.Normally,the expression Os06g0254600 is relatively low in flowers and leaves,whereas expression levels are relatively high in roots.When subjected to drought stress,it's expression are suppressed.Molecular weight of caleosins range from 15KD to 30KD.Caleosins have a significant feature that EF-hand domain in N-terminal can combine Ca2+,a central hydrophobic region and a security protein kinase phosphorylation sites in C-terminal.Caleosins's N-terminal and C-terminal are located in the cytoplasm,however central hydrophobic region are anchored in the phospholipid monolayer membrane.

Evolution

MEGA 4.0 software is used to build a phylogenetic tree.The figure shows that the caleosin genes of selaginella, moss and chlamydomonas are located at the bottom of the tree, indicating that caleosins originated in the early greenery.Rice caleosins ,arabidopsis caleosins and gymnosperms caleosins show two large different branches in evolutionary relationships This indicates that angiosperms and gymnosperms caleosin genes have at least two common ancestor. It has been found three kinds of gene replication mechanism:segmental duplication, tandem duplication and retrotransposition. Six caleosin genes in rice distribute on four different chromosomes.Os06g0254300,Os06g0254600, OsCLO-6 three genes are located to adjacent position of chromosome 6.Homology of Os06g0254300 and Os06g0254600reach to 92%,Os06g0254600 and Os06g0254700 is 49%.Indicating that Os06g0254300 and Os06g0254600 originated in a close repeated events, but Os06g0254700 and Os06g0254600 repeated earlier.
