Function

  • COM1/SAE2 is a highly conserved gene from yeast to higher eukaryotes. Its orthologs, known to cooperate with the MRX complex (Mre11/Rad50/Xrs2), are required for meiotic DNA double-strand break (DSB) ends resection and specific mitotic DSB repair events.
  • OsCOM1 plays an important role in promoting homologous synapsis and recombination in rice meiosis.

Phenotypic analysis

  • The mutant plants showed normal vegetative growth and development, and could not be distinguished from the wild-type plant, but were completely sterile. The male gamete fertility was examined by staining with 1% I 2 -KI solution.
  • Almost all pollen grains in the mutant plant were shrunken, with an irregular appearance, and could not be stained, compared with those from the wild type .
  • Furthermore, the mutant flowers did not set seeds when pollinated with the wild-type pollens, demonstrating that the mutation prevented both male and female fertility.
