Description In this project, Gene g6pd, trmt5 and dars2 conditional knockout mouse models were used to describethe core functions of trmt5 and dars2 in HSC metabolic regulation and maintenance; At the same time, using the human umbilical cord blood cd34+ system further elucidates the role of these genes in the maintenance of human HSCs and in vitro expansion. In addition, the project will collect umbilical cord blood HSCs under simulated physiological hypoxia condition, trying to protect HSCs from the stress of EPHOSS influence; Screening small molecular compounds that target PPP metabolism and mitochondrial metabolism to promote HSC amplification, and establishing effective HSC amplification system in vitro.
